β?葡聚糖对巨噬细胞增殖 胞内游离钙及cAMP浓度的影响
2011年3月25日 14:27 作者:郭兰芳【摘要】 探讨β?葡聚糖对RAW264.7细胞增生、胞内游离钙及cAMP浓度的影响。方法: 中性红比色法检测细胞生长, 酚试剂法检测细胞蛋白质合成, 动态检测β?葡聚糖对胞内游离钙和cAMP浓度的影响。结果: 微量β?葡聚糖可促进细胞生长及蛋白质合成;β?葡聚糖作用后3~5 min使胞内游离钙浓度升高,作用1 min 后使cAMP浓度升高, 并保持较高水平。结论: 胞内游离钙及cAMP是β?葡聚糖对RAW264.7细胞作用过程中重要的信使分子。 医学职称论文发表

[Abstract] Objective: To explore the effect of β?glucan on proliferation, intracellular calcium and cAMP levels of RAW264.7 macrophage cell line.Methods: Cell growth and proliferation were detected by neutral red colorimetric assay, protein synthesis by Lowry′s test, and the concentration of cytosolic calcium and cAMP were determined continuously. Results: Cell growth and protein synthesis were enhanced by β?glucan; the concentration of cytosolic calcium was elevated 3~5 min after treatment with β?glucan cAMP level increased 1 min after the treatment and maintained at high level. Conclusion: Cytosolic calcium and cAMP were important signal molecules in evaluating effect of β?glucan on RAW264.7 cell line.

随着免疫抑制剂、激素、广谱抗生素的大量应用,器官移植技术的广泛开展、各种导管的广泛应用及肿瘤治疗的广泛应用,真菌感染尤其深部真菌性医院感染日益增多。 β?葡聚糖是真菌细胞壁含量最高的多糖。本研究以小鼠巨噬细胞RAW264.7为观察对象,观察不同浓度及不同时间的β?葡聚糖对RAW264.7细胞的直接影响, 特别是对细胞内第二信使传递的影响, 以探讨β?葡聚糖对RAW264.7细胞的作用机制。医学职称论文发表
1 材料与方法
1.1 试 剂
RPMI1640、胎牛血清购自Gibco公司;Fura?2 AM 购自中国科学院药物所;β?葡聚糖和EGTA购自Sigma公司;其余试剂均为进口分装或国产分析纯。
1.2 细胞培养
小鼠巨噬细胞RAW264.7购自中国科学院上海生命科学研究院。于37℃,5%CO2湿润环境中培养, 每3天更换1次培养液, 细胞生长汇合成单层细胞时及时传代, 常规培养液为含10%胎牛血清的RPMI1640。
