【摘要】 目的 观察低能量氦-氖激光血管内照射(ILIB)配合中药治疗对实验性糖尿病脑梗死家兔神经元cNOSmRNA的影响。方法 经四氧嘧啶造成糖尿病及光化学方法造成脑梗死的高脂家兔糖尿病脑梗死模型35只,分为模型组、单纯激光照射组、中药(糖脑络通方)治疗组、激光+中药组,检测其神经元cNOSmRNA 水平表达,进行组间比较及与正常组比较。结果 与模型组比较,单纯激光和中药组对cNOSmRNA水平影响不大,而激光联合中药组可显著降低模型家兔神经元紊乱的cNOSmRNA水平,减轻nNOS介导的神经损伤,更有利于梗死后的神经功能恢复。结论 ILIB联合中药糖脑络通方可显著降低模型家兔神经元cNOSmRNA水平的表达,减轻nNOS介导的神经损伤,从而达到治疗糖尿病脑梗死的目的。

Method 35 successfully modeled rabbits, after alloxian injection for diabetes and photochemical radiation for stroke, were randomized into four treatment group:control group, ILIB group, traditional Chinese herb medicines treatment group, and a group with compound treatment of ILIB and TCH. 7 unmodeled rabbits were made as the normal group. The cNOSmRNA level was comparedwith eath other. Results Compared with control group, ILIB group and traditional Chinese herb medicines treatment group can’t significantly rectify the disorderly cNOSmRNA, but E can. Conclusions Group E can effectively reduce the cNOSmRNA level and nervous injury, cure diabetes cerebral infarction. qi1.1 仪器 ZJC480型电脑多功能氦-氖激光血管内照射治疗仪,长春中吉光电公司产品。JLT-B冷强光透照仪,河北泊头市医用光电仪器厂产品。1.2 造模 新西兰大白兔53只,购自中国药品生物制品检定所[北京市实验动物许可证,京动许字(2000)第017号总062号],体重2.5 kg左右,适应性喂养2周后,纳入实验。动物称重后随机抽出7只作为正常对照组,其余参考文献[3]造模。家兔禁食8~12 h,经耳缘静脉注射新鲜配制的5%四氧嘧啶生理盐水注射液,初次剂量为120 mg/kg,选取空腹血糖不足11.1 mmol/L者以150 mg/kg剂量注射。

\4O5iZg;P9{[ [ 选取经2次造模后空腹血糖在11.1 mmol/L以上者,每只每日给高脂饲料60 g(含1%胆固醇,15%蛋黄粉,5%猪油)加普通饲料喂养。2周后制作脑梗死模型。脑梗死模型:再参考文献[4-5]用光化学方法造成脑梗死。在以3%戊巴比妥钠1 mL/kg体重经耳缘静脉麻醉后,将其头部固定在立体定位仪上,取家兔左侧颅骨中线与冠状缝向后向左各0.6 cm处为中心,纵行切开头皮2 cm。暴露颅骨,用牙科车床锯开1 cm×1 cm大小的骨窗后,经耳缘静脉按1 mL/kg体重注射虎红(Rose Bengal, 3.5%),2 min后用冷强光透照仪经直径为0.7 cm的圆形光导探头由骨窗照射10 min。术区撒青霉素粉适量,局部以牙托粉封闭骨窗,逐层缝合切口。
