珠子参多糖抗肝癌作用的实验研究
2010年8月23日 14:08 作者:lunwenchina【摘要】 目的观察珠子参多糖(Panax japlcus var Polysaccharide,PJPS)对小鼠H22肝癌移植瘤生长的抑制效应及其机制。方法分别建立BALB/c小鼠H22肝癌移植实体瘤模型及腹水瘤模型,将小鼠随机分为3组,即生理盐水组(阴性对照)、5-氟尿嘧啶组(阳性对照)、珠子参多糖组。给药结束后比较各用药组的抑瘤率、胸腺指数、脾指数、肿瘤组织细胞形态结构,肿瘤细胞超微结构、肿瘤细胞凋亡率、CD4+/CD8+值、外周血血清中血管内皮生长因子含量及生命延长率。结果珠子参多糖组的胸腺指数、脾指数、CD4+/CD8+值,外周血血清中血管内皮生长因子含量及生命延长率均高于5-氟尿嘧啶组,珠子参多糖组的抑瘤率及肿瘤细胞凋亡率低于5-氟尿嘧啶组,珠子参多糖组的部分肿瘤细胞有坏死现象及凋亡学形态变化。结论珠子参多糖对H22肝癌小鼠具有抑制肿瘤生长、延长生命的作用。提高机体细胞免疫功能,干扰肿瘤细胞周期,降低VEGF的表达可能是珠子参多糖发挥抗肝癌效应的机制。

Abstract:ObjectiveTo observe the anti-tumor activity of Panax japlcus var Polysaccharide (PJPS) on transplanted tumor H22 in mice and its mechanism.MethodsH22 transplanted solid hepatocarcinoma model and the ascites hepatocarcinoma model were established.The BALB/c mice were randomly divided into 3 groups:normal saline group(negative control group),5-fluorouracil group(positive control group),and PJPS group. After administration,the inhibitory rate of tumor,the thymus index ,the spleen index,tumor cell morphology,and ultrastructure,the rate of apoptosis of tumor cell,CD4+/CD8+ ratio,level of VEGF and the rate of life extension were determined. ResultsThe thymus index,spleen index,CD4+/CD8+ ratio,level of VEGF and the rate of life extension in PJPS group were higher than those of 5-FU group,but the inhibiting rate of tumor and the apoptosis rate of tumor cell were lower than those of 5-FU group.The tumor cells of PJPS group partly died . The tumor cell nucleus of PJPS group concentrated, and tumor cells got smaller. ConclusionPJPS can inhibit the growth of hepatoma carcinoma cells of H22 tumor-bearing mice. Improving the cellular immune function, interfering with tumor cell cycle and reducing the expression of VEGF may be the mechamisms of antitumor effect.
