【摘要】 目的 分析宁夏地区牛脑组织博尔纳病病毒(Borna disease virus,BDV)的感染状况,探讨阳性基因扩增片段与德国马源性标准病毒株He/80同源性。方法 采用逆转录聚合酶链反应(RT-PCR)、荧光定量巢式逆转录酶聚合酶链反应(FQ-nRT-PCR)分别检测宁夏地区145例健康牛脑组织中 BDVp40、BDVp24基因片段,对BDVp24阳性扩增产物进行基因序列测定,并应用BLAST和DNAsist 5.0 软件对测序结果进行分析。结果 145例牛脑组织中4例BDVp40、BDVp24均阳性,阳性率为2.759%,且阳性BDVp24 片段的核苷酸序列与He/80 同源性最高,所编码氨基酸序列相同。结论 宁夏地区牛脑组织中可能存在动物源性BDV的自然感染,阳性BDV p24核苷酸序列与He/80具有高度同源性,所编码的氨基酸序列相同。

|+l'dY2h,FHc0d/_$^|0 Abstract: Objective To investigate the natural infection of BDV in bovine brain tissue in Ningxia, and to explore homology between gene amplification fragments in positive products and the German horse-borne standard strain He/80. Methods BDVp40 and BDVp24 gene fragments were detected by Revorse Transcriptase Polymerase Chain Reaction and fluorescent quantitative nested reverse transcriptase-polymerase chain reaction (FQ-nRT-PCR) in 145 cases of healthy cattle brain tissue in Ningxia. Gene sequencing was Determined in the positive PCR products and the sequence was analyzed using BLAST and DNAsist 5.0 software.Results There were 4 positive cases in 145 bovine brain tissue samples and the positive rate was 2.759%. The analysis of gene sequence showed BDV p24 in bovine brain tissue from Ningxia was homophylic with strain He/80,and their amino acid sequence remained the same. Conclusion BDV infection may exist in Ningxia bovine brain tissue.BDV p24-positive and Strain He/80 nucleotide sequence are highly homologous.

d/X:M-l]\k&N)i 博尔纳病病毒(Borna disease virus,BDV)是非分节段、单负链RNA病毒,具有非细胞溶解性、强嗜神经性特点,能感染从鸟到灵长类的广泛动物种类并引起以中枢系统功能障碍为特征的临床表现[1]。研究表明,在我国黑龙江、新疆、重庆、宁夏等地存在BDV的自然感染[2-4]。本课题组前期研究证实在宁夏地区牛、羊外周血单核细胞(PBMCs)及病毒性脑炎病人外周血单核细胞和脑脊液中存在BDV的自然感染,与德国马源性标准病毒株He/80序列比对,具有高度同源性[5-6]。BDV侵入机体首先进入血液或侵入嗅觉上皮或某些神经末梢,继而通过血液系统或神经轴突物质转运进入神经系统,造成行为异常、脑实质和脑膜的炎性细胞浸润,疾病特异性的抗原在边缘系统神经元中积聚,引起致死性中枢神经系统损害[7]。BDV 感染时外周血PBMCs 检测的阳性率与脑组织的阳性率是否一致呢?故本实验对宁夏健康成年牛脑组织BDVp40、BDVp24两个高度保守序列进行检测。
