【摘要】 目的: 研究积雪草酸(asiatic acid,AA)对人肝癌细胞株(HepG2)的抑制作用,并探讨其与线粒体之间的关系。方法: 采用MTT法检测AA对HepG2细胞增殖的影响,倒置显微镜观察细胞形态的变化;荧光探针MitoTracker检测线粒体数目;荧光探针JC?1检测线粒体膜电位;荧光探针DCFH?DA检测胞内活性氧(reactive oxygen species,ROS)含量;ATP试剂盒检测胞内ATP水平;RT?PCR和Western Blot法分析电压依赖性阴离子通道(voltage?dependent anion channel,VDAC)表达量的变化。结果: AA能够剂量依赖性抑制肿瘤细胞株的增殖,30 μmol/L AA作用细胞24 h后细胞突起消失、胞体皱缩、变圆。AA能够使HepG2细胞线粒体膜电位下降,胞内ROS含量增加,降低胞内ATP水平,50 μmol/L AA能够增加VDAC蛋白的表达量。结论: AA抑制肿瘤细胞增殖的作用与线粒体有关,线粒体VDAC参与人肝癌细胞凋亡的调控过程。

中国论文网+ET(LOs+o7i1\*g/m H'kGe:yT(g z$k1J0 [Abstract]Objective: To investigate the proliferation inhibition of asiatic acid(AA) on the human hepatoma cell line(HepG2) cells, and to study its potential correlation with mitochondria.Methods: The effect of AA on proliferation of HepG2 was tested by MTT, morphology of tumor cells was observed under inverted microscope; the mitochondrial mass was described by the fluorescence intensity of MitoTracker probe, mitochondrial membrane potential was detected with fluorescent probe JC?1, intracellular reactive oxygen species(ROS) content was detected with DCFH?DA,intracellular ATP level was measured by ATP Assay Kit, and the expression of voltage?dependent anion channel(VDAC) on mitochondrial outer membrane was determined by RT?PCR and Western Blot. Results: AA inhibited the proliferation of tumor cell lines(HepG2) dose?dependently, and cells rounded and shrank after treated with 30 μmol/L AA for 24 h.Mitochondrial membrane potential depolarized, intracellular ATP level decreased and ROS content increased dose?dependently after treating with AA, and VDAC expression was up?regulated by 50 μmol/L AA. Conclusion: AA exerted anti?tumor effects, which was related with mitochondria mediated death, and VDAC was involved in the cell death pathway.

S0J)R;u%[6J{2o0Cz+y Z3O`'i-W0 积雪草酸(asiatic acid, AA),又名亚细亚酸,是我国传统中药积雪草的五环三萜类组分之一。经研究证明, AA具有对抗CCl4和D?GalN诱导的肝损伤[2, 3]、减弱化疗药物抗肿瘤的不良反应[4]、对抗神经细胞的氧化损伤[5]等作用。另外,也发现AA具有抑制黑色素瘤细胞增殖[6]、诱导直肠癌细胞凋亡的作用[7]。本研究探讨了AA对肝癌细胞株HepG2增殖的抑制作用及其与线粒体之间的关系。
