免疫正常Wistar大鼠皮肤型孢子丝菌病的观察

【摘要】 【目的】 观察免疫正常Wistar大鼠皮肤型孢子丝菌病的发病过程,探讨其模型建立的可行性及实验条件。【方法】 将Wistar大鼠分为实验组(60只)和对照组(6只),在每只实验组Wistar大鼠背部皮肤选取两个接种部位:右背部及尾根部;制作申克孢子丝菌株SUMS0382的孢子悬液,并调整菌悬液浓度至孢子数为1 × 108 /mL;在实验组Wistar大鼠的每个选取部位皮下注射孢子悬液0.1 mL,造成实验性感染;对照组相应部位则注射8.5 g/L NaCl溶液0.1 mL。接种后12周内每日观察大鼠活力、体质量、皮损数量及直径,并于第2周、4周、8周分别取皮损组织行真菌培养及组织病理。【结果】 皮损在感染后第6天时开始出现,皮损数量及大小均在感染后第2周末达高峰。感染1周后,33%(20/60)的大鼠于接种部位出现皮损,感染2周后,90%(54/60)的大鼠于接种部位出现皮损,尾根部的皮损数目远大于右背部皮损数目。91%(49/54)大鼠的皮损表现为结节。19%(10/54)大鼠的皮损呈现反复的过程。皮损组织真菌培养及组织病理均符合皮肤型孢子丝菌病的改变。【结论】 免疫正常Wistar大鼠皮肤型孢子丝菌病的发病过程反映了人类发病的自然病程,免疫正常Wistar大鼠皮肤型孢子丝菌病模型的建立是可行的。 农业论文发表

Abstract: 【Objective】 To observe the development of cutaneous sporotrichosis in immunocompetent Wistar rat and investigate the feasibility and experimental conditions for its animal model establishment. 【Methods】 Sixty Wistar rats were inoculated with 0.1 mL of a suspension of 1 × 108 cfu/mL conidial inoculum from the Sporothrix schenckii strain SUMS0382 by subcutaneous route at two site: the right back and the root of tail. The control group was inoculated with 0.1 mL of 0.85% saline. Then activity, weight, number, and diameters of lesions of animals were observed daily for twelve weeks. Fungal culture and histopathology examination of tissue of lesions were conducted at the 4th week, 8th week, and 12th week after inoculation. 【Results】 Lesions were observed at the 6th day post-inoculation and reached to the peak at the end of the 2nd week. 33% of rats and 90% of rats were observed lesions at the inoculation site at the 1st week and 2nd week, respectively. Lesions of rats were mainly (91%) nodules and some of (19%) rats showed repeat courses. Fungal culture and histopathology examination of tissue of lesion confirmed the disease as cutaneous sporotrichosis. 【Conclusions】 Natural course of human cutaneous sporotrichosis was reflected in the Wistar rat and the establishment for immunocompetent Wistar rat model of cutaneous sporotrichosis was feasible.农业论文发表
孢子丝菌病是由申克孢子丝菌引起的皮肤、皮下组织及其附近淋巴系统亚急性或慢性感染,在极少数免疫力低下的人群可引起系统播散。在免疫力正常的人群中,皮肤型孢子丝菌病是最常见的感染类型,其病程长,治疗费用昂贵,皮疹常发生在身体暴露部位,严重影响患者的美观和社会生活[1]。国内外学者通过不同方法造成免疫抑制实验动物的系统型孢子丝菌病[2-5],以研究申克孢子丝菌菌株的毒力差异和系统型孢子丝菌病的致病机理,而关于最常见的皮肤型孢子丝菌病的动物研究少见。本研究首次利用免疫力正常的Wistar大鼠观察研究了皮肤型孢子丝菌病的发病过程,证实了建立免疫正常动物皮肤型孢子丝菌病模型的可行性,为进一步研究皮肤型孢子丝菌病的发病机理提供了基础。农业论文发表
